Malaria causes considerable morbidity and mortality among young children in malariaendemic areas worldwide. Anti-malarial antibodies are crucial determinants ofPlasmodium falciparum infection outcome. In this study the levels of circulatingimmunoglobulin-G antibodies specific to crude Plasmodium falciparum antigens weredetermined by an indirect enzyme-linked immunosorbent assay in a hospital based casecontrolstudy, of children with varying malaria severity living in the rural area of Gedarifeastern Sudan and El-Obied west of Sudan. The aims were to investigate ifimmunoglobulin-G subclasses correlated with the clinical presentation of the disease andother malariometric indices among children exposed to seasonal and unstablePlasmodium falciparum transmission. The levels of circulating interleukin-10 and thegenotypes of the single nucleotide polymorphism at position -1082 of IL-10 gene werealso investigated in relation to malaria severity. The results revealed that individuals with severe malaria had significantly lowerprevalence and levels of IgG1 and IgG4 as compared to those with uncomplicatedmalaria, suggesting a protective role for these molecules against severe malaria. Thelevels of IL-10 were lower in severe malaria patients as compared to those who presentedwith uncomplicated malaria, but no significant association was found between thespecific alleles or genotypes in the -1082 A/G polymorphic site and any particular formof clinical malaria. The present data argue for protective roles for IgG1 and IgG4 antibodies and IL-10against severe malaria, but find no evidence supporting significant effect of the differentalleles or genotypes at position (-1082A/G) of the IL-10 gene on protection from SM.

Results of studies that are based on the entire population cannot be applied on only a particular class of the society that possesses specific characteristics. The results of such studies will be inaccurate because of the negative influence of certain elements of society on the study group or non-importance of some elements of the society that can be neglected. Some conditions can be placed on the population so that the non-important part of the society can be trimmed if that part is outside the interest of the researcher. However, placing such conditions on the community, the data would not follow the same distribution of entire community. Hence, we need to find the distributional characteristics of the truncated data including the probability density functions and estimation of the parameters for the truncated distributions. This can be done for specific class distributions. In this thesis we discuss the distributional properties of and make statistical inference on the parameters of some truncated distributions among the exponential family such as the normal distribution, the Gamma distribution, and the Geometric distribution. Such probability distributions have different applications especially in economics and environmental applications. Therefore, we find the distributional properties and the estimates of the parameters of the normal distribution, the Gamma distribution, and the Geometric distribution. Finally, simulation studies were conducted to check the results obtained in this thesis.

Marketed antibacterial drugs suffer several problems, such as bitter taste and low stabilitywhich lead to patient incompliance. Prodrug technology for solving such problems isextremely exciting. Based on previously reported density functional theory calculations, amoxicillin ProD 1-2 and cephalexin ProD 1-2 were designed and synthesized. For theintraconversion of both antibacterial prodrugs the kobs and t1/2 values in different mediawere calculated from the linear regression equation obtained from the correlation of logconcentration of the residual prodrug versus time. At constant temperature and pH thehydrolysis reaction for the above mentioned prodrugs displayed strict first order kinetics asthe kobs was quite constant and a straight line was obtained. Kinetic studies in 1N HCl, pH2.5 and pH 5 were selected to examine the intraconversion of both prodrugs to their parentdrugs. The acid-catalyzed hydrolysis of the prodrugs was found to be much higher in 1NHCl than in pH 2.5 and pH 5. The experimental t1/2 values of amoxicillin ProD 1 in 1NHCl, pH 2.5 and pH 5 were 2.5, 7 and 81 hours respectively and for cephalexin ProD 1 in1 N HCl and pH 2.5 were 2 and 14 hours respectively. In contrast, t1/2 values of amoxicillinProD 2 in 1N HCl and pH 2.5 were 8 and 44 hours respectively and for cephalexin ProD 2in 1 N HCl was 6 hours. On the other hand, at pH 7.4, the four prodrugs were quite stableand no release of the parent drugs was observed. At pH 5 the hydrolysis of the prodrugswas too slow. The four antibacterial prodrugs were found to be bitterless. The bitter tastemasking by the prodrugs is believed to be via altering the ability of the drug to interactwith bitter taste receptors.
