Effect of Antiasthmatic Drugs on Cu-Zn Superoxide Dismutase and other Parameters in Hilla City

The study was conducted on asthmatic patients who were treated with inhaled salbutamol [patients (n=41), males=21, females=20] and asthmatic patients who were treated with Montelukast drug [patients (n=37), males=19, females=18], who were reviewing the center of allergies and asthma in Hilla city, for the period from January 2013 to the first thirty-June 2013. Dr. Raad Faanwkh and Dr. Abeer Al-Khfaagy participated in the selection of patients, and healthy subjects [control (n=39), males=19, females=20]. Serum was used to assay antioxidant enzyme activities, while erythrocytes (packed cells) were used to separate and purify the Cu-Zn superoxide dismutase enzyme.
The results of the study are as follows:
The study showed a significant decrease (P<0.001) in the concentration of total antioxidant capacity (TAC), and Cu-Zn superoxide dismutase activity in the sera of patients with asthma who used inhaled salbutamol or montelukast treatment compared with healthy controls. There was no difference in total antioxidant capacity (TAC) and the enzyme Cu-Zn SODs in the sera of asthma patients who used inhaled salbutamol compared with asthma patients who used the treatment of Montelukast.
Significant decrease in the SOD activity and TAC in the sera of patients with asthma (P<0.001) compared with the healthy group. The purified SOD had a specific activity of 2800 U/mg of protein (healthy), 2431.45 U/mg protein for inhaled salbutamol patients, and 2229.2 U/mg protein for treatment with montelukast drug patients, and gave a single band on polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulfate (SDS). Each of its subunits had a molecular weight of about [(18500 Daltons, control), (18300 Daltons, asthmatic patients)].
Serum levels of Zn, Cu, and Se were significantly different (p<0.001 for all) in patients when compared with healthy subjects. There was a significant decrease (p<0.001) in copper and selenium concentrations and a significant increase (p>0.05) in zinc concentration in the sera of asthmatic patients who used inhaled salbutamol and Montelukast when compared to those of the control group. Highly positive correlation (significant) between Cu and Cu/Zn (r=0.72), p value at the 0.01 level (2-tailed); highly negative correlation (significant) between Zn and Cu/Zn (r=0.65), p value at the 0.01 level (2-tailed); while weak negative or positive correlation (insignificant) between other parameter levels in group one. Highly positive correlation (significant) between Cu and Cu/Zn (r=0.64), p value at the 0.01 level (2-tailed); highly negative correlation (significant) between Zn and Cu/Zn (r=0.57), p value at the 0.01 level (2-tailed); positive correlation (significant) between Cu and TAC, total protein, Se (r=0.34, 0.33, 0.37) respectively, p value at the 0.05 level (2-tailed); while weak negative or positive correlation (insignificant) between other parameter levels in group two. Highly positive correlation (significant) between Cu and Cu/Zn (r=0.73), p value at the 0.01 level (2-tailed); highly negative correlation (significant) between Zn and Cu/Zn (r=0.58), p value at the 0.01 level (2-tailed); while weak negative or positive correlation (insignificant) between other parameter levels in group three.
These results indicated that oral Montelukast had some undesirable effect on copper status than inhaled salbutamol.
