Immunological and molecular determination of inflammatory cytokines (IL-1B , IL6 , IL10 and INF-gamma) for diagnosis of breast cancer

Breast cancer is the malignant tumor that forms from the uncontrolled growth of abnormal breast cells. It usually affects tissues involved in milk production (Ductal and lobular tissues). It is the most common malignancy in women and itremains one of the greatest health threats facing women around the world as we enter the 21st century. The total number of breast cancer patients involved in this study were (80) individual taken from Basrah oncology center in Basrah province, The age of patients range from 27-76 years and (80) individuals considered as control group after they were checked and confirmed to be free from any chest or any other health problems , The data of all patients were used to built a general classification model for breast cancer. The system offer an effect summary that examine each demographic factors that have an importance across multiple responses. The study showed the minimum number of breast cancer patients was found amongthe age groups 20-29 years (2.5%),followed by age group 30-39 years (12.5% ,above 40-49years (31.25%), the age group 50-59 years (32.5%) and >60 years (21.25%). P-value <0.0163.The residence of patients with breast cancer was found in central 45 (56.25%), followed by in peripheral 35 (43.75%), with P-value = 0.1008. The effects of various demographical factors effecting patients with breast cancerstarted with various marritalstatus that show the number of Singlepatients 3(3.75%), followed by married 71(88.75%) and divorced 6 (7.5), Theclinical features, that patients using surgery was 72(90.0%) and who do not the surgery 8(10.0%), Then comes the other factor the Chemotherapy, the patients with breast cancer who take the chemo therapy 77(96.25%) and who donot take chemotherapy 3 (3.75%) with P = 0.089, and the last factor metastasis the patients who have metastasis 23 (28.75%) and the patients not have 57 (71.25%). There were significant differences between BMI of patients with breast cancer (P=0.01), and show the majority of patients belong to overweight group 36 (45.0%) followed by obese patients 14(17.5%) (P-value = 0.0150). The data was recorded in stages of the breast cancer which notice the Stage of IIA was (25.00%) appeared in 22 breast cancer patient, followed by stage IIB was (23.86%), stages IA and IIIC were 2(2.27%) for each, and they are followed by stage IB 1(1.14%) , stage IIIA 19(21.59%), stage IIIB 3(3.41%) and stage IV 10 (11.36%) there were significant difference (P<0.001).The levels of interleukin IL-1βpg/ml among patients with breast cancer and control groups in various age group that show the age group (20-29) years (2.87 pg/ml) followed by (30-39) years (3.485 pg/ml) , continued with (40-49) years (4.168 pg/ml) , (50-59) years (4.733 pg/ml) and ( >60) years (5.049 pg/ml) with P-value = 0.03669 .The levels of interleukin IL-6 Pg/ml among patients with breast cancer and control groups in various age group that show the age group (20-29) years (4.69pg/ml) followed by (30-39) years (4.97 pg/ml) , continued with (40-49) years (6.30 pg/ml) , (50-59) years (6.28 pg/ml) and ( >60) years (6.65 pg/ml) with P-value = 0.01599.The levels of interleukin IL-10 pg/ml among patients with breast cancer and control groups in various age group show the age group (20-29) years recorded (2.18 pg/ml) followed by (30-39) years (22.15 pg/ml) , continued with (40-49) years (24.16 pg/ml) , (50-59) years (26.33 pg/ml) and ( >60) years (26.87 pg/ml) with P-value= 0.0042. The levels of IFN-γ pg/ml among patients with breast cancer and control groups in various age group that show the age group (20-29) years recorded concentration of (3.4 pg/ml) followed by (30-39) years (5.31 pg/ml) , continued with (40-49) years (5.90 pg/ml) , (50-59) years (6.48 pg/ml) and ( >60) years (6.72 pg/ml) with P-value = 0.0042.PCR amplification results were done on the extracted DNA and confirmed by using gel electrophoresis which appear as compact DNA separated bands which were results from the accurate and specific binding between the target DNA template and its specific primer, These bands viewed under UV imaging system and appear as illuminating orange bands due to ethidium bromide stain, only bands with expected molecular size 249bp were observed. IL-6 specific size 408pb. IL-10 specific size 295pb. IFN-γspecific size 262 pb.The sequence of the nucleotide of interleukin gene 25 microliters of each sample of the PCR product with the Primers of the nucleotide of IL1β , IL6 , IL10 and IFN-γ genes were sent to Macro-gene in the Korea and after obtaining the results all the results were compared directly with the nucleotide of the nucleotide of IL1B, IL6, IL10 and IFN-γ genes available in the internet (http: NCBI Reference Sequence) by computer program (Bio-Edit Pro. version: 7.0.0). The results were registered as anew recorded genes in NCBI under accession numbers (LC644661, LC644662, LC644663, LC644664, LC644665, LC644666, LC644667 and LC644668). Which is available on the website (ncbi.nlm.nih.gov ). The study able to record a genes in NCBI these genes have a multiple mutation and has a specific location on DNA chromosome.
