Etude du pouvoir antifongique des extraits de plantes sur Fusarium spp. de l’orge en conservation

L’objectif de ce travail a visé la recherche d’un biofongicide à base d’extraits aqueux de plantes médicinales contre trois isolats de Fusarium spp. qui attaquent les graines conservé pour les cultures des céréales hydro-thermiques utilisée en Algérie. On a choisit les plantes utilisé dans le domaine d’agro-alimentaire qui ont une pouvoir antifongique : Eugenia caryophyllata, Lavandula stoechas, Cinnamomum zeylanicum, Punica granatum, Aloysia citrodora, Pistacia lentiscus et Allium sativum. Les extraits aqueux ont été préparés selon deux méthodes d’extraction : la décoction et l’infusion à une concentration de 150mg/ml. Un screening phytochimique a été réalisé pour mettre en évidence quelques métabolites secondaires des plantes. L’évaluation de la teneur en polyphénols dans chaque extrait avait pour objectif de confirmer leur pouvoir antifongique vis-à-vis des isolats de Fusarium spp. étudiés. Le screening phytochimique a révélé la présence de saponosides dans l’extrait aqueux à base de Pistacia lentiscus. Le dosage des polyphénols a montré une teneur plus élevée dans les extraits aqueux de l’Aloysia citrodora et la Punica granatum avec des teneurs de 74,8mgEAG/gMS et 64,5 mgEAG/gMS respectivement. Parmi les 7 plantes étudiées, l’extrait aqueux préparé à base d’Eugenia caryophyllata par la méthode de décoction a montré une inhibition complète de la croissance mycélienne et de la sporulation (100%) des deux isolats de Fusarium spp. F1 et F2 selon la méthode contact direct.Il est intéressant de rechercher des autres extraits efficaces pour tous les isolats étudiés de Fusarium spp. Il sera aussi préférable de poursuivre notre recherche par l’étude de l’activité antifongique in vivo de l’extrait à base de clous de girofle et identifier les molécules responsables de l’effet fongicide en vue de formuler et appliquer le biofongicide en culture hydroponique de l’orge fourrager
(For the past years micro-organanisms have slowly developed a resistance to chemical pesticides, giving rise to the necessity to find a natural fongicide based on the aqueous plant extracts to inhibit their development and reduce their pathogenicity. Our study is based on the anti-fungal activities of the aqueous extracts of Eugenia caryophyllata, Lavandula stoechus, Cinnamomum zeylanicum, Punica granatum, Aloysia citrodora, Pistacia lentiscus and Allium sativum against the three isolated samples of Fusarium spp. , which attack Barley cultivated using the hydroponic system. We prepared the aqueous extracts using two methods: decoction and infusion. The direct contact method was used for the study of the anti-fungal activity of the aqueous extracts and a dilution was made for the extracts that showed a high inhibitory effect. The dilutions were: 37.5mg/ml, 75mg/ml and 112.5mg/ml. An organoleptic and a physiologic analysis was conducted on these extracts after. We also measured the quantities of the polyphenolic content in the plant materials using the Folin Ciocalteuagent and very high values were retained for the extacts of Punica granatum and Aloysia citrodora at 74.8mgEAG/gMS and 64.5mgEAG/gMS respectively. This analysis was done to study correlation between the concentration of polyphenols in plant extracts and their inhibitory effects against pathogens. A high presence of saponosides was noted in the extracts of Pistacia lentiscus after some phytochemical tests. Amongst the seven plantes that were selected for this study the aqueous extracts of Eugenia caryophyllata caused a total inhibition of the mycellial growth and sporulation for the isolated samples of Fusarium spp. at a concentration of 150mg/ml using the direct contact method. It would be beneficial and interesting to conduct this research in vivo using the Eugenia caryophyllata and to identify the molecules responsible for this high inhibitory effect so that we can reformulate and apply this biofongicide in the hydroponic system)
