Studies on Bacillus Species metabolites and their bactericidal effect against Staphylococcus species

الملخص

The present study was carried out to isolate and identify the differenttype of Bacillus sspfrom different sources.And totested their metabolites asbactericidal to StaphylococciThirty two Gram positive bacilliwere isolated from different sources.The main source was contaminantsin bacteriological cultures.The isolates were screened for their production of metabolites againstStaphylococcus (Staphylococcus aureus, Staphylococcus epidermidis andStaphylococcus saprophytycus).Three of these isolates were positivefor production of metabolites thatcompletely inhibitedStaphylococcus growth.The three positive isolateswereBacillus cereus, Bacillus lichniformis and Bacillus subtilisThe metabolites were isolated and partially purified from thesespecies in order to study their chemical and physical properties. The result ofchemical and physical properties of the active metabolites filtrates revealedthe presence of an extracellular thermosensetive (the metabolite degradingby high temperature ) ,but these metabolites could be kept at roomtemperature for long time .In order to establish the inhibitoryactivity unit different metabolitefiltrates dilutions were tested against Staphylococcus spp.The inhibitoryactivity unit which is higher dilution of metabolite filtrate that inhibits invitro the Staphylococcusgrowth Was1x10-1viiiFrom plate assay to establish the bacteriostatic or bactericidal activityof metabolite filtrate, the result revealed that the metabolites havebactericidal effect against staphylococcus spp.The metabolite filtrate stability testagainst enzymes revealed that themetabolite filtrates were denatured byprotease enzyme
