Production, Characterization and Partial Purification of Cellulolytic and Pectinolytic Enzymes from Three Selected Fungi

Cellulose and pectin are carbohydrate polymers, much abundant as an agricultural residues or wastes. They are resistant to enzymatic cleavage in most animals. Researches around the world were therefore directed towards finding methods for hydrolyzing these compounds. One such approach is enzymatic hydrolysis. Three fungi (Trichoderma viride, Asperigillus niger and Penicillum digitatum) are used for production of cellulase and pectinase enzymes. This study was aimed at producing characterizing the cellulase and pectinase enzyme systems of above fungi under laboratory conditions. Two methods (reducing sugar and viscometry) were used for measuring enzymes activities. Cellulose, carboxymethylcellulose, pectin and sodium poly pectate compounds were used as substrates. The production of cellulases and pectinases enzymes of the three fungi was investigated in culture media. A. niger and T. viride gave maximum production of cellulase enzyme after one week. While, P. digitatum gave it after two weeks. T. viride and P. digitatum gave maximum production of pectinase enzyme after two week and A. niger gave it after one week. On the other hand, the maximum temperature for cellulase enzyme activity was found at 50°C for A. niger. The optimum temperature for pectinase enzymes of both A. niger and P. digitatum were detected at 40°C, while it was at 50°C for T. viride. The optimum pH for cellulase activity of A. niger was detected at 4.0 but it was 6.0 for T. viride. Concerning pectinase enzyme the optimum pH was found at 9.0 for A. niger. Purification by ammonium sulphate precipitation, the maximum cellulase enzyme activity was at 60% concentration for the three fungi, and maximum pectinase activity was at 80% concentration for both fungi A. niger and P. digitatum. By using gel electrophoresis, three bands were showing cellulolytic activities. However, five bands were showing pectinolytic activities. This study showed the importance of fungi as sources of enzymes and recommends that more studies must be done in the field of biotechnology to produce more glucose from natural product by using fungi.
