الملخص
Maytenus dhofarensis is a celastraceous herb naturally found in the southern region of Oman, where it is locally known as "Serbit." Ethnobotanical reports suggest that consumption of the plant’s fruits by goats induces illness characterized by episodes of shivering. Although M. dhofarensis is not traditionally recognized as a medicinal plant in Omani herbal practices, other species of the Maytenus genus are known globally for their therapeutic properties. Due to the lack of prior phytochemical studies on this species, this study aimed to explore M. dhofarensis for structurally novel compounds and assess their bioactivities against kinases involved in cell survival and proliferation signaling pathways. In this investigation, hexane and chloroform extracts from the fruits of M. dhofarensis were chemically analyzed, leading to the isolation of seven compounds: Monoacetyldiglyceride (1) Triacylglyceride (2) Palmitic acid (3) Hydroxylated monoacetyldiglyceride (4) Dihydro-β-agarofuran-type sesquiterpene pyridine alkaloid (5) Dihydro-β-agarofuran-type sesquiterpene pyridine alkaloid (6) Alkenyl isothiocyanate (7) All seven compounds are newly reported isolates from M. dhofarensis. Their structures were elucidated through comprehensive spectroscopic analyses, including IR, 1D and 2D NMR, UV, and ESI-MS. The relative configuration of compound 5 was further established using Density Functional Theory (DFT) calculations in conjunction with NMR data. In addition, analysis of the seed oil using GC-MS with DMOX derivatization revealed the presence of hydroxylated fatty acids, including palmitic acid, stearic acid, and oleic acid (18:9). Notably, compound 5 demonstrated potential anticancer activity by inducing morphological changes and partially inhibiting colony formation in MCF-7 breast cancer cells. Molecular pathway analysis showed a significant downregulation of phosphorylated Akt (p-Akt) in MCF-7 cells upon treatment with compound 5, suggesting a targeted mechanism of action.
