Mosquitoes are an important example of the evolutionary success of insects, which contribute significantly to biodiversity and biomass. Species represent about 3450 species, many of which pose a major medical and economic hazard to humans and animals. Medical females occupy a dominant position among all arthropods In addition to the discomfort and anxiety caused by human comfort and sleep due to its tinnitus and painful tingling, it is unique in that it is the only carrier of many bacterial, viral and parasitic pathogens while feeding on human and animal blood Because of the medicinal and veterinary importance of mosquitoes, and the evolutionary success it has shown, and being an important vector for many pathogens (viral, bacterial, parasitic), and bacterial communities residing in these vectors, which may provide opportunities for manipulation and thus reduce the efficiency of mosquitoes For bacterial, viral and parasitic pathogens. So, The aim of this study were Classification of medicinal insects (mosquitoes) under the Culicinae family in Aleppo based on global and local taxonomic keys.Identification of the types of bacteria transmitted by mosquito species studied using microbiological methods. Confirmation of some isolated bacterial species using molecular methods. Study of bacterial germs associated with studied mosquitoes. Identification of some types of viruses transmitted mosquito types studied using immunological methods .A total of 2524 samples of Culicinae mosaics were collected from the study sites and were categorized in five species belonging to three species: Cx. pipiens, Cx. quinquefasiatus, Cx. the ileri, Cs. longiareolata, Ae. dorsalis A difference was found in the species in the study group, which was the species Cx. pipines The most abundant species in Aleppo were 86.09% of the total number of mosquitoes in the study areas. However, its abundance differed from one region to another depending on the natural and geographic factors prevailing in each site Culex quinquefasciatus came in second place with 8.66%, Culisetalongiareolata5.27%, Aedesdorsalis0.32%, Culextheileri the least abundant species, 0.16% And Cx.pipiens were collected in high numbers in each of the cellars of the University, Al-Ramousaand New Aleppo, while the Aedesdorsalis and Cx.theileri were collected from the Kawakibi area and the university vaults. Fourteen bacterial species were isolated from 12 genotypes, which were followed by three germs: Proteobacteria, Firmicutes and Actinobacteria, divided into five groups: Gammaproteobacteria, Alphaproteobacteria, Bacilli, Clostridia and Actinobacteria. The results of the isolation indicate that the bacterial flora of the mosquito females. The total and studied in this study is a complex mixture of bacterial species, dominated by Gamma proteobacteria, with 57.54% gram negative bacteria, the percentage of gram-negative bacilli was 50%, the percentage of gram positive bacillus was 14.29%, the percentage of gram-positive bacillus was 28.57%, while the gram-negative percentage was 7.14%.Pseudomonasaeruginosaand E. coli were also isolated from the controlled samples, indicating that these bacteria are the natural flora of the gastrointestinal tract of mosquitoes. Some bacterial species were documented in molecular biologic methods using traditional and cultured PCR technology, using specialized prefixes for genes specific to each bacterial type. Escherichiacoli was documented using PCR technology using UIDA (uidA 858 / uidA 1343). The results of the PCR analysis showed that the length of the inflated uidA was approximately 480 bp, (UidA1047 / uidA1232) on the previous PCR products, the length of the segment was 180 bp, and thus the bacterial type was documented. Staphylococcus aureus was confirmed and documented using the nuclease (St1-St2) genes. After the electrophoresis of the PCR amplification products and DNA profiles, the length of the DNA fragment was amplified by 270bp.Klebsiellapneumoniawas confirmed using the two prefixes (K2 -R / K2 -F). The results of the extracted DNA transcription showed that the length of the amplified DNA segment was 650bp.Staphylococcusepidermidiswas confirmed and documented using the two indices (TetK-F / TetK-R). The results of the PCR analysis of the electrolytic process of the DNA derived from human staphylococcus aureus showed that the length of the DNA fragment from the TetK gene amplified by the PCR reaction The previous two were approximately 350bp.The ure-F / ureR of the gene ureR were used to confirm Proteus. mirabilis. The results of the PCR analysis showed that the length of the DNA sample from the ureR gene amplified by the PCR reaction and using the two previous genotypes was approximately 220 bp .Thep-Ab-ITSF/p-Ab-ITSRof 16S-23Swere used to confirm Acinetobacter baumannii .The results of the PCR analysis of the extractedthe DNA using the two previousmethods of electroplating process showed that the length of the amplified DNA was 200 bp.Wolbachiapipientiswas also investigated for female mosquitoes using PCR technology as it did not grow on normal culture media using the W-Spec-R, W-Spec-F, 16S rDNA genes. PCR on DNA extracted from female mosquitoes studied using W-spec-F, W-spec-R, specimens of Wolba chiapipientis. Wolbachia pipientis DNA was found in four insect cultivars of 80% belonging to Culexpipiens, Culexquinquefasciatus, Aedesdorsalis, while Culisetalongiareolata was not found, and the presence of these bacteria in male mosquitoes was found in Culexpipiens, Culexquinquefasciatus using PCR technology and using the two prefixes .The presence of viruses (WNV / WEE / SLEV)and Alphaviruses in female mosquitoes was investigated from the study areas using immunosuppressant Vec-Test based on the principle of immunoglobulin, no viral species were recorded from the Alphaviruses group. While the West Nile virus virus was recorded for the first time in Aleppo and Syria, in two mosaics, one belonging to type Cx.pipiens from the Ramousa region, and the second from the type Cx.quinquefasciatus of the Adonis region .The existence of this virus is linked to many things, such as the presence of the host carrier in abundance and abundance (mosquitoes), and the presence of host buffalist (birds, especially crows), and the presence of this virus in many neighboring countries such as Palestine, Jordan, Turkey and many Arab countries, and low health awareness, Economic life, climate change and urbanization, these things can lead to the emergence of this virus and many other diseases
