Estrogen is a key hormone that helps protect women’s hearts by supporting healthy blood flow and reducing inflammation in the arteries. It also assists in controlling blood pressure and maintaining balanced cholesterol levels. These effects contribute to a lower risk of heart disease in women before menopause. When estrogen levels drop after menopause, the risk of developing cardiovascular problems increases significantly. The aims of the current study was to investigate the effect of estrogen depletion by VCD on the Antioxidants (Glutathione GSH and Nitric oxide NO)oxidants (Malondialdehyde MDA and Peroxy nitrite ONOO-)cardiac biomarkers (Troponine I and Creatin Kinase-MB) estrogen receptors α gene expression and estrogen receptors β gene expression,ECG P,QRS,T wave ( ms) P,QRS.T wave (mv), QT interval (ms), ST interval (ms), R-R interval (ms) ,heart rate ,histopathological exam (heart,ovary) Ten female rats are randomly divided into two groups each group with five animals, the first set was the control group injected with normal saline, and the second group received 4-Vinylcyclohexene Diepoxide (VCD) 80 mg/kg BW/daily for 2 weeks as 4-Vinylcyclohexene Diepoxide (VCD) group started from 1 /10/2024 to 15/10/2024 in the animal house in College Of Veterinary Medicine/ University Of Kerbala. After collecting samples at the end of the experiment showed the results showed a significant decrease in the serum level of estrogen for the VCD group compared to the control group, increases in feed intake, water intake, body weight and heart weight with increase in serum level of cardiac troponin-I , creatin Kinase-MB, ONOO- , MDA, with a significant decreases in NO and GSH. Also there was down regulation in ERα gene expression in heart tissue ,and there was a upregulation in ERβ gene expression compare with control group. .A decrease in waves of (P,QRS,T) time with significant increase in voltage of waves (P,QRS) there was no significant difference in T wave compare with control group ,also show that were a significant (p ≤ 0.05) increase in heart rate in the VCD group due to shortened in QRS waves. The histopathological exam of the heart in VCD group showed shows myocardial degeneration with vaculoation of cardiomyocytes, necrosis and infiltration of neutrophils with inflammatory cell and interstitial hemorrhage, the aorta of VCD group increase thickness of vessels wall due to fibrosis with mononuclear cells infiltration.while in the ovary showed an degeneration of primary follicles with thickness of granulosa with marked vacuolation of interstitial cells with interstitial hemorrhage and sever fibrosis with sever congestion and edema of blood vessels and thickness of endothelium basement membrane with sever eosinophils and fibroblast infiltrationIn conclusion the recurrent study revealed that estrogen depletion by VCD caused disturbance in the oxidants /antioxidants damage and Cardiac Troponin and Nitric Oxide , creatin kinase with positive correlation between estrogen-ERα and negative correlation between estrogen-ERβ, there is a decrease in the level of estrogen in the group that was injected with the VCD, and this depletion causes an increase in the heart voltage.
