- In order to elucidate the complex regulatory network that controls nitrogen fixation in Rhizobium etli CNPAF512 strain, we focussed our work on the regulation of the expression of nif Agene, coding the key transcriptional activator of the nif/fixgenes in rhizobia. In fact, NifA is the masterregulator of nitrogen fixation and plays a central role in signal transduction by switching on and off the genetic cascade of nif/fixgene activation, ensuring expression of the nitrogen fixation apparatus during symbiosis. Analysis of the effects of 5’ progressive deletions in the promoter region of nif Agene in R. etli and assessment of their functionality at the level of expression allowed us to show marked differences between the different deletions and also depending on the growth conditions, thereby indicating that specific symbiotic regulatory mechanisms are involved. Interaction of the nif A promoter with putative regulatory proteins has been investigated by electrophoretic mobility shift assays as well, clearly showing that a histone-like protein interacts in a specific manner with the promoter region pnifA. Entire genome screening of R. etli strain allowed us to identify several clones that could contain genes coding histone-like proteins. In fact, in one of these clones, we could individualize a gene, coding a histone-like protein we tentatively named HURe. Several in vitro and in vivoexperiments were then realized to characterize the properties of this protein. Therefore, we could show that HURe protein displays distinct binding characteristics to DNA substrate, as compared to Escherichia coli HU protein. These particular properties individualize this protein and indicate that, similarly to other histone-like proteins, HURe interacts in astructure-specific, rather than sequence-specific, manner with DNA, and particularly suggests that it could have a different function in the cell
