- Urtica Dioïca L. known by the vernacular name “Horaig” is a medicinal plant of the Urticaceae family, which grows in the Mediterranean region and widely used in traditional medicine. The aim of the present study is to evaluate the effect of the nature of the extraction solvent: ethanol and hexane, as well as the extraction method: maceration on the yield, content of bioactive substances, the polyphenols, and flavonoids)and on the estimation of the antioxidant activity of the aerial part of the nettle harvested in the region of Ouadhias Wilaya of Tizi-Ouzou. In this work, two extracts were prepared, from the aerial part of this plant, two solvents (water and water-ethanol mixture50:50 v/v) with a new method (hot maceration with stirring) for the extraction of polyphenolic substances were tested on the plant, as well as the calculation of the contents of these compounds. Studies on antioxidant and biological activity were also carried out on the extracts obtained to determine the effectiveness of Urtica Dioïca with respect to oxidizing agents and bacterial microorganisms. The results obtained show that the extraction solvent and the extraction method remarkably affect the yields and the secondary metabolite content as well as the antioxidant and antibacterial activity. The best yield was acquired with water as extraction solvent of 9.46%. Likewise, water is the most effective for the extraction of phenolic compounds (18.75 mg/g), as well as for flavonoids (9.575 mg/g). The evaluation of the antioxidant power which was carried out using the DPPH free radical scavenging method and showed good antioxidant activity at 59.8% at the concentration of3.24 mg/ml. The aqueous extract is endowed with the highest reductive power than other extracts. This study demonstrates that the plant studied has considerable antioxidant activities. These result scan be a starting point for applications of this plant in the prevention of various diseases
