- The research presented in this dissertation Magister fall within the themes of our research team, Nutrition and Metabolism in the laboratory LBPO de la Faculté des Sciences Biologiques de l'USTHB. In this work, we are interested in studying the establishment and evolution of the metabolic syndrome (MS) induced by a hypercalorique (RHC) and its consequences at the level of the aortic wall of Psammomys obesus and the effect of an oral antidiabetic agent metformin on the development of this syndrome. In a first part, we are interested in the ontogenesis of the aorta of this animal model particularly well suited to this kind of study at various stages of development from the fetal status at birth, the pre pubescent then to adulthood. The histological study of the different aortas at different stages of development, has shown that age is a determining factor in the morphological changes and vascular remodeling induced by RHC and significant changes in biochemical such enrichment collagen and GAG, which may be interpreted as risk factors for atherosclerosis. In the second part of this brief, we studied the effect of RHC on adult Psammomys because in its natural habitat this animal does not develop neither diabetes nor obesity or atherosclerosis. Our assays showed that after 3 months of RHC, 100% of the animals become obese, 70% of them grow, in addition, hyperglycemia, hyperinsulinemia, hypertriglyceridemia and hypercholesterolemia. All these phenomena are growing up to 6 months of RHC, and in 30% of them, a clear SM moves. The animal is unable to regulate his blood sugar level after 6 months and most of them become insulin-requiring. On the histology of the aorta affected Psammomys presented to RHC shows a thickening of the endothelium as a major disruption of the elastic tissue and with thinning and rupture of the strong growth of collagen. The media thickens with high GAG deposits. The CMLS the media invade the intima in response to the increasing infiltration of circulating monocytes. The CMLS are bloated, they appear larger because of the presence of edema in the arterial wall and increased clusters of vasa vasorum in the adventitia. All these phenomena worsen with the duration of the RHC. In the third part of our study, we tested the effect of an oral antidiabetic agent of the family of biguanides: metformin (Met) on the development of MS in diabetic Psammomys made after 3 months of RHC. We found, after 6 months of treatment with metformin (dose of 5 to 25 mg / kg of animal dose administered daily according to blood glucose level), a slight decrease in body weight (4 g on average after 3 months of treatment followed by a return to normal after 6 months of treatment), a considerable decrease in blood glucose and insulin levels which can be explained by the action of Metformin which has the effect of favoring the use glucose by tissues, especially by skeletal muscle and the inhibition of hepatic néoglycogenèse and, accordingly, a significant decrease in cholesterol and triglycerides. We have finally shown through the MM and QSAR relations, modeled the action of the Met on glucose, L-Lysine and the complex [Glucose-L-Lysine] that the Met had the immediate effect, by a phenomenon of competitive reaction, block all active sites of glucose when it is in the bloodstream in its very reactive aldehyde.
