Rheumatoid arthritis (RA) is an inflammatory joint disease responsible for the joint destruction. It is a disease that involves genetic, environmental and hormonal factors. Genetic susceptibility to complex diseases has been intensively studied during the last decade. The HLA-DRB1 locus is known as the most important genetic contributor for the risk of RA, but is not sufficient to drive autoimmunity. Thus, other genetic and environmental factors are involved. Over 30 non-HLA single nucleotide (SNPs) predisposing to disease have been identified in Caucasian and Asian populations. However, few studies have focused on the study of architecture of RA in the Algerian population. Our work has focused on a study type case/control with a view to identify the potential susceptibility genes for RA in the Algerian population, by analyzing some candidate genes. The study focused on 343 RA patients and 345 healthy subjects. The SNP of PTPN22 gene (rs2476601), PADI4 (rs224034), IL23R (rs11209026, rs1343151 and rs10489629) IL17F (rs2397084 and rs763780), IL-17A (rs2275913) and IL2/IL21 (rs6822844) were by real time chain reaction (RT-PCR) using technology (Applied 7500). Our results showed no association of the variants of PADI4, IL23R and IL17 genes to the development of RA in our population. However, we have demonstrated that the minor allele T of PTPN22 rs2476601 is associated with rheumatoid factor (RF) positive disease. This suggests that this could be involved in the activation of B cells; hence, in the production of in RA. Also, an association between the two SNPs of IL23R gene (rs1343151 and rs10489629) and the production of the different of RF (IgG, IgM and IgA) was revealed. Finally, we were able to highlight the strong association of the SNP IL2/IL21 (rs6822844) to RA. Indeed, our data showed a protective effect of the minor T allele of rs6822844 ; whereas the major G allele appeared to be susceptible. The -phenotype analysis of variants revealed an association of the two IL23R (rs1343151 and rs10489629), the IL17Fand IL2/IL21 with the clinical parameters of RA. The Replication of this study on a larger cohort of patients and controls could clarify the role of these SNPs in the of RA in our population. Also, the study of the functional consequences of the of PTPN22, IL23R (rs1343151 and rs10489629) and IL2/IL21 (rs6822844) genes would be interesting to understand the mechanisms by which they might contribute to the pathology.
