Contribution to the study of nutritional influences on immunity in Psammomys obesus and Rattus norvegicus-Diabetic therapeutics based on extracts from seeds of Citrullus Colocynthis In laboratory rats made diabetes by streptozotocin In the first chapter, the standard laboratory diet administered to sand rat (Psammomys obesus) induces the following physiological and immunological changes: hyperglycemia and hypercholesterolemia involving mainly the free fraction of cholesterol, with an elevation of high-density-lipoprotein levels and a decrease in B and T splenic lymphocyte proliferation in the presence of different mitogens PHA-P, Con A and LPS. These results demonstrate the important modification that could be induced in sand rat by the standard laboratory diet as compared with natural diet, and thus the sand rat (P. obesus) appears to be an interesting model for studies on experimental diabetes mellitus. In the second chapter, we assessed the effects of insulin, cytokines and cyclosporin A on mitogen-stimulated T lymphocytes of streptozotocin-induced diabetes mellitus in Wistar rat. Diabetes induced by streptozotocin in Wistar rat was followed by hyperglycemia and hyperlipidemia. Lymphocyte proliferation, whether resting or stimulated by T-cell mitogens, was always lower in diabetic rats as compared to the control group, and the decreased T lymphocyte proliferation of diabetic rats could be restored by addition of insulin. Cyclosporin A (CsA), a known immunosuppressor, curtailed the insulin- and IL-1-potentiated T lymphocyte proliferation both in normal and diabetic rats. However, the CsA failed to influence the mitogen-stimulated T-cell proliferation. These results suggest that IL-1 and insulin act on T cells via CsA-sensitive pathway. Furthermore, the T cells of normal rat contained higher basal intracellular free calcium, [Ca 2+]i concentrations than those of diabetic rat. Insulin-stimulated [Ca 2+]i concentrations were more pronounced in normal T cells than the diabetic cells. Addition of cytokines (IL-1 and IL-2) to T cells recruited higher [Ca 2+]i concentrations in diabetic group as compared to the control group. These results suggest that the [Ca 2+]i homeostasis of T cells and their functioning, altered in diabetic rats, might contribute to immunosuppression in this disease. In the third chapter, we try to evaluate the antihyperglycaemic effect of the aqueous extract, saponins, flavonoids, total alkaloids and chloroformic glycosidic extract from seeds of colocynth (Citrullus colocynthis L. Shard) “Handal”, family of cucurbitaceae, in normal and streptozotocin diabetic rats. This plant was used as traditional remedy for treatment of diabetes in folk medicine in the Maghreb and the Middle East. First, toxic effect of these compounds was determined by the determination of the LD50. Results showed an antihyperglycemic effect of all extract studied. The different compounds of the colocynth did not lowered significantly blood glucose levels in fasted normal rats.
