Association of Body Mass Index and Obesity Related FTO gene (rs17817449, rs8043757 and 8063946 ) polymorphisms in Obese Adults, Gezira state, Sudan 2017

Obesity is a common and serious medical problem. It is a major cause of morbidity and mortality worldwide. Fat mass and obesity associated FTO gene was identified as most polygenic factor associated with body max index. The current study was designed to investigate the associations between the FTO rs17817449 and the FTO rs8043757 polymorphisms in intron one and obesity. Sixty participants (45 as case, 15 of them as control) were involved in this study, their weight and height were measured to obtain Body Mass Index (BMI) and structured questionnaire was completed to assess, eating behavior and physical activity. DNA was extracted from all 60 whole blood sample by guanidine chloride method. The PCR was carried to amplify the FTO rs17817449 region .Normal sequencing was used to detect the point mutation. The sequence was analyzed using Finch TV and Bio edit programs. The results of this study indicate the presence of FTO rs17817449, and FTO rs8046757polymorphisms.There was significant statistical association between FTO rs17817449, FTO rs8046757polymorphisms and the main diet (p = 0.02). There was no association with physical activity levels and these polymorphisms (p=0.08). Novel FTO8063946 was found in one case which needs further study to detect it is effect.
