The present work deals with the use of the supercritical fluid extraction to obtain oils from two chosen local natural plants, namely seeds of Peganum harmala and Plantago ovata. The extraction process was carried out under different experimental conditions of pressure, temperature and grain size. Using Box Behnken, the optimum seed yield of Peganum harmala was 15.001% at 300 bar, 55°C and a grain size of 0.5 mm, whereas for Plantago ovata seeds, it was 11.109% at 240 bar, 60°C and 0.25 grain size with. The biological activities (antioxidant, antibacterial and antifungal) of these extracts were also evaluated. DPPH radical scavenger was used to assess the antioxidant activity. The optimal values for the IC50 and the TPC were 172.99 μg/mL and 79.04 mg GAE/g E, respectively at an extraction pressure of 100 bar, a temperature of 35°C and a particle size of 0.9 mm. However for the TFC, an optimal value of 7.10 mg QE/g E was obtained at a pressure of 170.7 bar but still at a temperature and a particle size of 35°C and 0.9 mm, respectively, while no activity was recorded for the seeds of Plantago ovata, in contrast to extracts obtained by means of Soxhlet extraction using methanol, where the results were estimated at 90 mg/ml and 35 mg/ml for the seeds of Peganum harmala and Plantago ovata, respectively. As for the antibacterial activity, the ability of 15 Peganum harmala seed and 15 Plantago ovata plant extracts to stabilize 4 types of bacteria in addition to the candidate yeast was tested. As for antifungal activity, seven mushrooms were tested. Different results were obtained depending on the bacteria and the used fungi. Moreover the different results were related to the adopted experimental conditions. The results obtained by Soxhlet extraction were much better than those extracted by supercritical carbon dioxide, despite the advantages brought by supercritical carbon dioxide. This drawback encouraged the development of a molecular design strategy of supercritical fluids, using the group contribution concept. New five supercritical fluids (CF4, CF3CF3, CF3CH3, CH2NCF3and CF3CF2CH3), were tested and showed much better solubilities of two chosen solutes (Climbazole, Irgacure) in supercritical CO2at given ranges of pressure and temperature, contrary to a third solute (Triclocarbon) where only the proposed CF3CF2CH3was slightly better than supercritical CO2.Finally another approach was proposed to optimize the composition of the extraction according to a particular compound directly related to a particular targeted activity of property. This remains to be refined further to end up with an important tool for the obtention extracts to measure.
