Hepatitis B is a global infectious disease, that seriously threatens human health. There are mainly interferon and nucleoside analogs treatment for Hepatitis B in the clinical practice, which has certain therapeutic effects on Hepatitis B. Still, its many harmful effects and drug resistance are increasingly reported. The traditional screening methods of anti-HBV activity take time and effort and it is difficult to obtain accurate screening results. So, it is urgently needed to discover and develop novel anti-HBV drugs, preferably natural product compounds, which might have high anti-HBV efficiency, and low toxicity with novel antiviral mechanisms. In this computational study, marine-derived drug-like natural products were used to inhibit Hepatitis B virus replication. Three novel compounds (L-2-hydroxyproline phenylalanine, glycine-L-proline, and 4-hydroxyproline phenylalanine) were selected and subsequently subjected to molecular docking using Molecular Operating Environment software (MOE). There were docking analyses for L-2-hydroxyproline phenylalanine that have shown better molecular characteristics and binding free energies than glycine-L-proline and 4-hydroxyproline phenylalanine. Additionally, it which significant molecular interactions with the active site residues of Hepatitis B virus targeted protein. Furthermore, the analyses have shown two-hit compounds against each targeted protein showing stable behavior and higher binding affinity and key residual molecular interactions. Despite the limited computational studies, we anticipate that the identified hits could substantially influence the development of novel therapeutics against Hepatitis B in the future.
