The current study was conducted to investigate the bacterial causes of eye infection, which include inflammation of conjunctivitis, eyelid infection (blepharitis), and dacryocystitis at different age groups. One hundred ninety-one swabs were collected from the eyes of infected people in addition to 191 blood samples of people who reviewed Al-Hilla Teaching Hospital, Alnoor Child Hospital, Babylon Maternity and Paediatric Hospital for the period November 2011 to May 2012. Detection of Chlamydia’s role in eye infection was carried out by using Chlamydia IgG and IgM specific diagnostic kits. Only one case of 42 eye infection samples showed positive results for anti-Chlamydia IgM. Culture investigations showed 176 positive bacterial growths out of 191. The bacterial isolates included 119 Gram-positive and 57 Gram-negative isolates. The Gram-positive isolates included Staphylococcus aureus (38.06%), Staphylococcus epidermidis (10.22%), and Streptococcus sp. (19.31%), while the Gram-negative isolates included Escherichia coli (22.7%), Neisseria sp. (2.84%), Klebsiella pneumoniae (5.12%), and Pseudomonas aeruginosa (1.70%). Measurement of the total number and differential count of white blood cells by routine methods showed an increase in W.B.C. count in patients compared with controls, especially in the age groups 31–40 and 61–70 years. The study also dealt with the detection of cytokine concentrations. IL-6 was increased at this concentration, especially in the age group 41–50 pg/ml, reaching 8.52, and Interleukin-12 was increased in its concentration in patients compared to controls, particularly at ages less than one year, reaching 13.45 pg/ml. While interferon gamma was higher in the 41–50 years group, statistical analysis did not show significant differences between patients and control groups. PCR assays were performed to identify the presence of some genes related to pathogenicity in bacteria responsible for inflammation of the eye. Staphylococcus aureus represented the largest causative agent of inflammation of the eye. The study revealed the presence of certain genes in some bacterial isolates, including the Staphylococcus enterotoxin A gene, which appeared in large proportions in the study isolates (21 out of 24) at a rate of 87.5%, while in the case of the Staphylococcus enterotoxin B gene, its existence appeared in a few isolates (2 samples out of 24) at a rate of 8.3%. Moreover, the toxic shock syndrome toxin gene was disclosed but did not show any infection rate. This indicated that the isolated bacteria in this study lacked this gene.
