الملخص
This study included 70 swabs collected from burned patients of both sexes, males and females of different ages attending the burn unit at Al-Hilla Teaching Hospital in Al-Hilla city during a period of three months from October to December 2011. The results revealed that 100% of samples showed positive results for bacterial culture. The results indicated that 26/70 (37.01%) of samples were Pseudomonas aeruginosa and 44/70 (62.9%) revealed other types of bacteria, which included 23/70 (32.9%) Staphylococcus spp. and 21/70 (30%) Escherichia coli. The susceptibility of Pseudomonas aeruginosa towards six antibiotics included in this study by Disk Diffusion Test showed that all P. aeruginosa isolates (100%) exhibited resistance to Rifampin, Gentamycin, Pipracillin, and Tobramycin, while they were sensitive to Meropenem and Amikacin. In addition, P. aeruginosa revealed remarkable susceptibility toward natural products, Garlic Vinegar, but it was not affected by Garlic Oil. The minimum inhibitory concentration (MIC) for the six antibiotics using the Pouring method was carried out. The results showed that P. aeruginosa isolated from patients was resistant to most of the antibiotics used in this study. MIC values were 64 μg/ml for Amikacin and Meropenem, 256 μg/ml for Rifampin, while no inhibition zone was observed on the bacterial culture tested with Gentamycin, Pipracillin, and Tobramycin. To detect the response of Multi-Drug Resistant isolates of P. aeruginosa towards a combination of antibiotics with each other using the Well Diffusion method, it was shown that Meropenem and Amikacin revealed high potent activity against the tested organism when combined with other antibiotics, indicating synergy and additive action. However, Amikacin and Tobramycin revealed an inhibition zone of 43 mm in diameter. Meropenem combined with Pipracillin and Amikacin showed an inhibition zone of 43 mm and 37 mm in diameter, respectively. According to these results, the six antibiotics were employed for in vivo experiments, since they were used for treatment of experimentally infected animals alone and in combination. The response of experimentally infected animals in recovery from experimentally burned infection was remarkable when combined antibiotics were used for treatment in comparison to the use of single antibiotics, which indicated that a synergy reaction was observed, and the results were in accordance with the Well Diffusion method.
