ترجمة الملخص
1165 isolates of bacteria and yeasts from different foodstuffs were identified using API technique, including 342 isolates of Gram-negative bacteria belonging to Enterobacteriaceae family (69 Escherichia coli, 53 Enterobacter cloacae, 23 Klebsiella oxytoca,26Proteus mirabilis,11 Citrobacter koseri,8 Salmonella. spp, 7 Morganella morganii, 14 Hafnia alvei, 9 Pantoea. spp and 9 Serratialiquefaciens),the accuracy rate of API 20 E and API ID 32 E identification systems for Enterobacteriaceae species were 62% and 79% respectively. The genus Pseudomonas)54 Pseudomonas aeruginosa ,27 Pseudomonas fluorescens,18Pseudomonas stutzeriand14Pseudomonas luteola),the accuracy rate of API 20 NE system was 37%. The 609 isolates of Gram-positive belonging to the genus Bacillus(66Bacillus subtilis,24Bacillus cereus,7Bacillus anthracis,28Bacilluspumilus, 19Bacillus licheniformisand 31Bacilluslentus(, the accuracy rate of API 50 CHB and API 20 E system together to identify Bacillus species was 72% and for Staphylococcus(65Staphylococcus aureus, 39Staphylococcus epidermidis,24Staphylococcus haemolyticus,21Staphylococcus hominis, 13Staphylococcuslugdunensisand 8Staphylococcus cohnii) the accuracy rate of API STAPH and API ID 32 STAPH systems were 71% and 77%, respectively. The bacilli and cocci belonging to lactic acid bacteria (29Lactobacillus paracasei,14Lactobacillus rhamnosus,9Lactobacillus brevis, 46Lactobacillus plantarum, 7 Lactobacillus fermentum, 25 Lactococcus lactis, 23 Leuconostoc mesenteroides, 60 Enterococcus faecalis, 44Enterococcus faeciumand 7Streptococcus thermophilus(, the accuracy rate of API 20 STREP, rapid API ID 32 STREP and API 50 CHL systems to identify Lactic acid bacteria species were 43% , 75% and %54 respectively. Foryeasts (26Saccharomyces cerevisiae,33Candida kefyr,40Candida krusei,36Candida lusitaniae, 16 Candida lipolytica, 29 Rhodotorula mucilaginosa,9Rhodotorula minuta,7Debaryomyces etchellsii/carsonii, 10Zygosaccharomyces. sppand1Candida holmii), The accuracy rate of API 20 C AUX and API ID 32 C systems were 32% and %35 respectively. The Real-Time Polymerase Chain Reaction technique was used to identify some of the lactic acid bacteria. The fatty acid composition of bacteria and yeasts cells was analyzed by means of GC-MS. The results revealed the differences in fatty acid profiles both qualitatively and quantitatively in different bacteria and yeasts species, that enabled use to identify them accurately and to consider fatty acid profiles a genetic fingerprint as a tool for identification. The Gram-negative bacteria were characterized by the presence of saturated and monounsaturated straight-chain fatty acids, hydroxy and cyclopropane fatty acids, while Gram-positive bacteria (except for lactic acid bacteria) were characterized by the presence of saturated and monounsaturated straight-chain and branched fatty acids. Moreover, lactic acid bacteria characterized with the presence of cyclopropane fatty acids and the absence of branched fatty acids. The yeasts characterized with the presence of saturated, monounsaturated and polyunsaturated straight chain fatty acids. The results showed the superior of GC-MS technique on API system in terms of accuracy, rapidity and cost efficiency. The accuracy rate of the latter technique was 83%, 90%, 86%, 82%, 80% and 90% for Enterobacteriaceae, Pseudomonas, Bacillus, Staphylococcus, Lactic acid bacteria and yeasts respectively. The effect of growth conditions (temperature, age of colony and media composition on S. aureus, and the absence of Tween 80, aeration, pH and NaCl concentration) on fatty acid profiles were studied. The results revealed that the Lb. paracaseifatty acid composition of bacteria changed according to growth conditions, so the conditions of growth were standardized for bacteria and yeasts when identified via GC-MS. Moreover the results proved the accuracy of identification of Staphylococcus species using GC-MS when MSB medium was modified. This research was performed in the laboratory of National Commission for Biotechnology in the period of 2010-2014. The aim of this research was to identify some bacteria and yeasts based on their cellular fatty acids qualitatively and quantitatively by means of Gas Chromatography Mass Spectrometry technique (GC-MS).
