الملخص
Les métalloprotéinases du venin de serpent (SVMPs) sont les principaux composants responsables des dommages tissulaires locaux observés après envenimation. Cette étude décrit la purification et la caractérisation d’une nouvelle SVMP dermonécrotique, la CcD-II, isolée à partir du venin de C.cerastes. Cette protéinase de 39,9 kDa est douée d’une importante activité protéolytique vis-à-vis de plusieurs composants de la MEC notamment le collagène IV. La caractérisation de la réponse inflammatoire locale associée à le dermonécrose a également été décrite, révélant une réaction inflammatoire exacerbée qui pourrait contribuer à l’amplification des dommages tissulaires induits par les SVMPs, impliquant les MMPs et le NO.

- يشكل التسمم الناتج عن لدغ الافعى سيراستاس سيراستاس (Cerastes cerastes) مصدر اعراض صحية خطيرة مقترنة بالتهاب معتبر. تعد الميتالوبروتينات (SVMPs) من مكونات سم الافاعي الرئيسية والمسؤولة عن التلف الموضعي للأنسجة بما في ذلك النزيف وتقرح الجلد. في إطار دراسة سم هذا النوع من الافاعي تم استخلاص ووصف الخصائص البيولوجية والبيوكيمائية لمركب ميتالوبريتين جديد مسؤول عن تقرح الجلد وينتمي الى القسم II.ويكون هذا البروتين المسمىCcD-II من سلسلة واحدة من متعدد بيبتيد ذات وزن جزيئي يقدر ب 39,9kDa. كشف ال CcD-IIعن قدرة عالية لتحليل الكازيين، الجيلاتين، الكولاجين IV، اللامينين والفيبرينوجين مما يثبت مساهمته المعتبرة في الاضرار الفيزيوباتولوجية الناتجة عن تدخله في الاتصال الخلوي.علاوة على ذلك فان هذه الدراسة اهتمت أيضا بوصف الاستجابة الالتهابية الموضعية المصحوبةعمتقرح الجلد، فكشفت عنانتاج هام في MPO ،NOMMPs ،IL-6 وTNF-αبعد التسممنأجئاتنلاتنّيب.SVMPsالمستخلص من سم سيراستاس سيراستاس بما في ذلك ال CcD-IIتلعب دورا في الاستجابة الالتهابية من خلال تدخلها المباشر في افراز وتفعيل ال pro-MMP-9و تفعيل الpro-MMP-2.وبالإضافة الى ذلك فقد اثبت في هذا العمل ان الاستجابة الالتهابية يمكن ان تساهم في التأثير الباثولوجي الموضعي لل SVMPsباشراك ال MMPs وNOفي تضخيم الاضرار الموضعية للأنسجة بعد التسمم. ان التقرح النسيجي الناتج عن التسمم بسم الافاعي عبارة عن عملية معقدة ناتجة عن تأثير ال SVMPsعلى سلامة وتلاحم الخلايا وعلى الاستجابة الالتهابية أيضا. ومن اجل فهم أفضل للآليات الدقيقة المتدخلة في هذه العملية، إجراء المزيد من التحاليل البنيوية والوظيفية يعتبر أمرا ضروريا. ان هذه الدراسة قد تساعد على شرح الملاحظات الإكلينيكية المسجلة في حالات لدغ الافاعي بما في ذلك العلاج المضاد للتسمم الذي اظهر ضعفا في الفاعلية، كما انها تطرح قضايا علاجية هامة متعلقة بالتسمم الموضعي. Envenomation by Cerastes cerastesviper is characterized by pathological alterations associated with a prominent inflammation. Snake Venom Metalloproteinases (SVMPs) are the main components responsible for local tissue damage including haemorrhage and dermonecrosis. This study describes biological and biochemical characterization of CcD-II, a novel dermonecrotic SVMP-II isolated from C. cerastes venom. This proteinase called CcD-II is a single polypeptide chain with a molecular weight estimated to 39.9 kDa. CcD-II exhibited a high proteolytic activity on casein, gelatin, type IV collagen, laminin and α-β-chains of fibrinogen, indicating thus its substantial contribution in the induced pathophysiological alterations by interfering in cell connectivity. Moreover, the characterization of the local inflammatory response associated with dermonecrosis has been also described in this study, showing significant production of MPO, NO, MMPs, IL-6 and TNF-α induced after envenomation. The results showed that SVMPs of C.cerastesvenom including CcD-II play a role in the inflammatory response by being directly involved in the expression and activation of pro-MMP-9 as well as in the activation of pro-MMP-2. In addition, it was shown in this work that the inflammatory response may contribute to the local pathological effect of SVMPs, by involving MMPs and NO in the amplification of local tissue damage induced after envenomation. Tissue necrosis occurring after viper envenomation is a complex process that is induced by the effect of SVMPs on cell integrity and adhesion and also on the inflammatory response. Further structural and functional analysis will be necessary to understand the exact mechanisms involved in tissue necrosis process.This study could help to explain the clinical observations that treatment by antivenom after snake bite is only minimally successful and raises important therapeutic issues regarding the treatment of local envenoming.
