Nouvelles voies de Synthèse des 1,4-dihydropyridines, polyhydroquinoléines et 1,4-benzoxazines

الملخص
Le travail entrepris à travers cette thèse entre dans le cadre du développement de nouvelles méthodes de synthèse de molécules d’intérêt biologique, dont l’objectif principale était de réaliser des réactions à composants multiple donnant lieu à des molécules originales.Cette thèse est composée de trois chapitres principaux: le premier et le deuxième ont fait l’objet de l’étude de nouvelles voies de synthèse des 1,4-dihydropyridines et des polyhydroquinoline via la réaction de Hantzsch, et la synthèse des tétrahydropyridines hautement fonctionnalisées respectivement et ce en utilisant deux nouveaux catalyseurs pour ces synthèses. Alors que le troisième chapitre contient deux parties : dans la première partie on a réalisé la synthèse d’une gamme d’amines secondaires qui ont servi comme produits de départ pour la deuxième partie, qui traite de la préparation d’une série de nouveaux dérivés de la 4-hydroxy-1,4-benzoxazine synthétisé, en une seule étape, via la réaction de Petasis. Dans le premier chapitre, nous avons décrit l’utilisation d’un catalyseur inédit dans la réaction multicomposants de Hantzsch. Il s’agit du t-BuOK qui est un catalyseur de type base,utilisé avec une quantité catalytique de 10 %mol il a conduit à des séries de 1,4-dihydropyridines et de polyhydroquinolines avec des rendements bons à excellents.Dans le deuxième chapitre, nous avons étudié la synthèse de tétrahydropyridines mais en employant un catalyseur de type acide de Lewis. Il s’agit d’un dérivé de triflate : le Bi(OTf)3, qui a montré une bonne activité catalytique envers cette synthèse et a conduit à l’obtention de molécules désirées avec de bons rendements.Finallement, dans la première partie du dernier chapitre nous avons réalisé la synthèse d’une série d’amines secondaire, via la réciton d’amination reductrice. Par contre, dans la deuxième partie de ce chapitre, nous avons préparé une série de 2-hydroxy-1,4-benzoxazines selon une réaction de type Petasis, où la réaction consiste en une cyclocondensation entre l’aminoalcool préalablement préparé dans la première partie, un acide boronique, et le glyoxal. Cette réaction nous a permis d’obtenir neuf nouveaux produits avec des rendements bons à excellents qui ont fait l’objet d’une publication. - The work undertaken through this thesis is part of the development of new synthetic methods leading to biologically interesting molecules, whose main objective was to carry out multicomponent reactions resulting in original molecules.This thesis is divided into three main chapters: the first and the second were the subject of the study of new synthetic routes to 1,4-dihydropyridine via Hantzsch reaction, and the synthesis of highly functionalized tetrahydropyridines respectively and using two new catalysts for these syntheses. However, the third chapter contains two parts: in the first part we realized the synthesis of a range of secondary amines which have served as the starting material for the second part, which deals with the preparation of a series of new derivatives 4-hydroxy-1,4-benzoxazine synthesized via the reaction Petasis.In the first chapter, we have described the use of a novel catalyst in the multicomponent Hantzsch reaction. The t-BuOK which is a basic catalyst was used with a catalytic amount of 10 mol% and led to a series of 1,4-dihydropyridine and polyhydroquinolines with good to excellent yields.Like in the second chapter, we studied the synthesis of tetrahydropyridine but using a Lewis acid catalyst. It is a derivative of triflates: Bi (OTf) 3, which showed good catalytic activity envres this synthesis and leads to the production of desired molecules in good yields.Finally, in the first part of the last chapter we have synthesized a series of secondary amines via reductive amination rection. However, in the second part of this chapter we have prepared a series of 2-hydroxy-1,4-benzoxazine in a Petasis type reaction, where the reaction is a cyclocondensation between the amino alcohol previously prepared in the first Part, boronic acid, and glyoxal. This reaction has allowed us to get nine new products with good to excellent yields, which were the subject of a paper.Keywords: 1,4-dihydropyridine, polyhydroquinoline, Hantzsch, t-BuOK, tetrahydropyridines, Bi(OTf)3, 4-hydroxy-1,4-benzoxazine, secondary amines, reductive amination, Patasis.
