Apport thérapeutique de l’essence aromatique du thym (Thymus vulgaris L.) dans la lutte-prévention des pathologies fongique et inflammatoire

L’objectif assigné à ce travail consiste à valoriser l’essence aromatique d’une plante à parfum, Thymus vulgaris L., en aromathérapie anti-infectieuse et/ou comme ingrédient actif dans la prévention des pathologies inflammatoires topiques.La composition chimique de l’huile essentielle (HE), déterminée par Chromatographie Gazeuse-Spectrométrie de Masse, a révélé la présence de 13 constituants. Le composé majoritaire est le carvacrol avec un taux de 83.8%, suivi par le p-cymène (8.15%) et γ-terpinène (4.96%).Le screening antifongique de l’HE a été accompli par plusieurs méthodes sur différentes souches mycéliennes (9 levures et 7 moisissures). En phase liquide, l’HE a exhibé une activité inhibitrice remarquable sur la croissance de toutes les souches étudiées avec des Diamètres des Zones d’Inhibition variant entre 53 et 85 mm. Cette huile est douée aussi d’un pouvoir fongicide à forte dose car toutes les souches ont été inhibées. En phase vapeur, de meilleurs résultats ont été obtenus notamment pour Candida tropicalis où une inhibition totale a été notée à la plus faible concentration. De plus, cette action inhibitrice est « dose-dépendante ». Une méthode quantitative, dilution en milieu gélosé, a été explorée afin de déterminer les Concentrations Minimales Inhibitrices (CMI). Toutes les souches levuriformes ont des CMI faibles, de l’ordre de 0.3 µL/mL, excepté Rhodotorula sp. (0.15 µL/mL)Sur le plan thérapeutique, le potentiel anti-inflammatoire topique de l’HE a été exploré in vivo. Par la méthode de l’œdème à la l’huile de croton, le calcul du pourcentage de réduction de l’œdème a révélé que les lots traités avec l’HE, à différentes doses (2.5, 0.25 et 0.05 mg/mL), ont présenté un taux élevé (65%-73%), statistiquement identique (p>0.05) à celui du contrôle positif. Cette activité a été aussi confirmée, à l’échelle cellulaire, par une étude anatomo-pathologique.En somme, les résultats obtenus laissent entrevoir des perspectives d’application pharmaceutiques de l’essence du thym commun dans la prévention-traitement des infections fongiques et des pathologies inflammatoires locales.

- The aim of this study was to enhance the essential oil of an aromatic and medicinal plant, Thymus vulgaris, as antimicrobial agent in aromatherapy and/or as an active ingredient in the prevention or management of topical inflammatory diseases. The chemical composition of Thymus vulgaris essential oil (TVEO) was determined by Gas Chromatography-Mass Spectrometry and revealed the presence of 13 compounds. Carvacrol was found to be the major component (83.8%), followed by p-cymene (8.15%) and γ-terpinene (4.96%). Antifungal screening of TVEO was assessed by several methods against various fungal strains (9 yeasts and 7 molds). In the liquid phase, TVEO exhibited a potent inhibitory effect a gains tall the fungal strains with Diameter of Inhibitory Zones (DIZ) varying between 53 to 85 mm. With the highest dose, TVEO was illustrated by a fungicidal power against all mycelial strains. In the vapor phase, better results were obtained in particular with the strains of Candida tropicalis where complete inhibition was observed at the highest concentration. Else, this inhibitory effect of TVEO was "dose-dependent." The Minimum Inhibitory Concentrations of yeast were determined by agar dilution method and revealed that MIC varied from 0.3 to 0.15 μL/mL for yeast strains. Topical anti-inflammatory potential of TVEO was also explored in vivo by the test of croton oil-induced ear edema. TVEO exhibited a potent anti-inflammatory effect at all doses (2.5, 0.25 and 0.05 mg/mL) which were statistically similar to the positive control. This activity was also confirmed at the cellular level by a histopathology analysis.
