Isolement et caractérisation de biosurfactants bactériens

الملخص
L’objectif principal de ce travail est de produire des agents de surface en cultivant deux souches de collection du genre Rhodococcus sur des huiles végétales, ainsi qu’un consortium bactérien et deux de ses membres, Ps. citronellolis KHA et S. marcescens LHA, sur le gazole, et de caractériser les substances isolées des milieux de culture. En parallèle, les capacités de dégradation par les bactéries sélectionnées de deux polluants potentiels, l’huile de tournesol résiduelle de friture et les hydrocarbures, ont été évaluées en vue d’une utilisation éventuelle dans la restauration de sites contaminés par ces substances toxiques. A cet effet, nous avons isolé par enrichissements successifs avec le gazole utilisé comme seule source de carbone, et ensuite identifié un consortium bactérien issu d’une boue active prélevée d’un bac de stockage d’ hydrocarbures de la raffinerie d’Alger. Ce consortium composé de sept souches a présenté des capacités à tolérer (et à utiliser) le gazole à des concentrations initiales supérieures à 15% (vol/vol), ce qui correspond à une concentration de 126 g/L. Les conditions de croissance les plus favorables du consortium bactérien ont été déterminées : le taux spécifique de croissance optimal est de μ=0,211±0,005 j-1 (j=jour) pour une concentration initiale en gazole égale à 25,2 g/L. Les isolats KHA et LHA ont été cultivés avec le gazole comme seule source de carbone à la concentration de 25, 2 g/L: le taux spécifique de croissance est de μ=0,194 ±0,050 j-1 pour KHA et μ=0,146 ± 0,001 j-1 pour LHA ; la culture des autres souches a fourni des résultats plus mitigés en terme de taux de croissance. Ces résultats suggèrent que le consortium présente une meilleure aptitude à utiliser le gazole. Les extraits bruts des cultures bactériennes ont présenté des propriétés physico-chimiques très intéressantes : l’activité émulsifiante de ces produits bruts, évaluée par la mesure du taux d’émulsion de Cooper & Goldenberg, a atteint la valeur de 78% pour le consortium, 75% pour KHA, 37% pour LHA, 63% pour 16LM USTHB et 32% pour la souche 12LM USTHB. Des analyses en InfraRouge, CPG et CPG-MS/IE ont permis d’identifier tous les agents de surface produits par les souches testées. Les deux souches 12LM USTHB et 16LM USTHB ont produit, respectivement, des glycolipides et des esters méthyliques et éthyliques d’acides gras. KHA a synthétisé des esters propyliques d’acides gras, et la souche LHA, des acides gras. Ces composés peuvent trouver des applications dans les industries pharmaceutiques et des cosmétiques, notamment. Compte tenu de leur pouvoir émulsifiant exceptionnel, les extraits bruts synthétisés par KHA pourraient être utilisés à des coûts raisonnables dans la récupération secondaire des huiles fossiles, sans qu’il soit nécessaire de les purifier.

- The main objective of this work is to produce biosurfactants by cultivating two strains of the Rhodococcus genus with vegetable oils, as well as a bacterial consortium and two of its members (KHA and LHA) on the diesel oil, and to characterize the substances isolated from the culture media. In parallel, the degradation capacities of the selected bacteria towards two potential pollutants, namely the residual sunflower oil of frying and hydrocarbons, were evaluated for a possible use in the restoration of sites contaminated by these toxic substances. For this purpose, we isolated by selective enrichments with the diesel oil used as the sole carbon source, and then identified, a bacterial consortium sampled from an active sludge of a crude oil storage tank located in the Algiers oil refinery. The consortium, which consists of seven strains, has exhibited good capacities to tolerate (and use) the diesel oil with initial concentrations higher than 15% (vol/vol), which corresponds to a concentration of 126 g/L. The optimum conditions for growth of the bacterial consortium were determined: the optimal specific growth rate reached the value of μ = 0,211±0,005 J -1 for an initial diesel oil concentration corresponding to 25, 2 g/L. The isolates KHA and LHA were cultivated with the diesel oil as the sole carbon source at the initial concentration of 25, 2 g/L. The specific growth rate reached the value μ = 0,194 ±0, 05 J -1 for the strain KHA and μ = 0,146 ± 0,001 J -1 for LHA; the crop of the other strains provided less interesting results in terms of growth rate. These results suggest that the consortium presents a better aptitude to use the diesel oil The crude extracts from the bacterial cultures presented physico-chemical properties very interesting. The emulsifying activity of the crude products, evaluated by the measurement of the emulsion index rate of Cooper & Goldenberg, reached the value of 78% for the consortium, 75% for KHA, 37% for LHA, 63% for 16LM USTHB and 32% for the strain 12LM USTHB. Infra-Red, CPG and CPG-MS/IE analyses made it possible to identify all the surfactants produced by the strains tested. Two strains 12LM USTHB and 16LM USTHB produced, respectively, of the glycolipides and methyl and ethyl esters of fatty acids. Taking into account their exceptional emulsifying capacity, the crude product synthesized by KHA could be used at reasonable cost in the fossil oils recovery, without it being necessary to purify it.
