الملخص

En raison de l'importance des plantes dans la fabrication de médicaments et de remèdes naturels, car elles contiennent des composés actifs efficaces pour éliminer les maladies, cette étude visait à isoler et à extraire le composé actif naringine des agrumes, plus précisément des écorces d'orange, et à évaluer ses activités biologiques in vitro et in silico. La naringine a été extraite des pelures par trempage dans du méthanol (solvant organique) avec un rendement de 3,5% (poids sec). Un test a été mené pour mesurer l'efficacité de la naringine dans chacune des activités suivantes (anticancéreux, antidiabétique, antibactérien), où les résultats ont montré une forte affinité entre la naringine et les molécules biologiques, avec un rapport de liaison de 1,84 x 10^4 M^-1 pour l'ADN et 1,09 x 10^5 M^-1 pour la BSA. Quant à l'effet antibactérien du composé, il a ensuite été évalué sur deux souches (Staphylococcus aureus, Escherichia coli), et d'après les résultats, il a montré une activité inhibitrice de la naringine sur les souches testées. Les résultats ont également montré l'efficacité du composé pour inhiber l'enzyme alpha-amylase responsable de la dégradation de l'amidon et de sa conversion en sucre, avec un taux d'inhibition de IC50 = 3,5 µg/ml. Afin de vérifier les résultats de laboratoire avec une image informatique, nous avons effectué le processus d'amarrage moléculaire afin d'évaluer la formation et la force de la liaison entre la naringine et les différents composés biologiques à travers le programme de simulation de modélisation moléculaire AutoDock, dans lequel les résultats ont montré la formation de liaisons hydrogène entre chacun des groupements du composé et les acides aminés des molécules participant à l'interaction. Des simulations de dynamique moléculaire ont été réalisées sur les résultats de l'amarrage moléculaire du complexe (naringine - alpha-amylase) afin de mesurer la stabilité thermodynamique à la fois du composé et de l'enzyme, les analyses ayant montré la formation de liaisons hydrogène, ainsi que la stabilité du composé et de l'enzyme et la flexibilité de son site actif.

Due to the importance of plants in the manufacture of medicines and natural remedies, because they contain active compounds that are effective in eliminating diseases. This study aimed to isolate and extract the active compound naringin from citrus fruits, specifically from orange peels, and to evaluate its biological activities by computer and in the laboratory. Naringin was extracted from the peels by soaking using methanol (an organic solvent) with a yield of 3.5% (dry weight). A test was conducted to measure the effectiveness of naringin in each of the following activities (anti-cancer, anti-diabetic, anti-bacterial), where the results showed a strong affinity between both naringin and biological molecules, with a binding ratio of 1.84 x 104M-1for DNA and 1.09 x 105M-1for DNA. As for BSA, then the antibacterial effect of the compound was evaluated on two strains (Staphylococcus aureus, Escherichia coli), and according to the results, it showed an inhibitory activity of naringenin on the tested strains. The results also showed the effectiveness of the compound in inhibiting the alpha-amylase enzyme responsible for the demolition of starch and converting it into sugar, with an inhibition rate of IC50 = 3.5 μg/ml. In order to verify the laboratory results with a computer image, we performed the molecular docking process in order to evaluate formation and the strength of the bond between naringin and the different biological compounds through the molecular modeling simulation program AutoDock,in which the results showed the formation of hydrogen bonds between each of the amino acids of the compound and the amino acids of the molecules Participate in the interaction.Molecular dynamics simulations were performed on the results of the molecular fusion of the complex (naringenin -alpha-amylase) in order to measure the thermodynamic stability of both the compound and the enzyme, as the three analyzes showed the formation of hydrogen bonds, as well as the stability of the compound and the enzyme and the flexibility of its active site
