A molecular Study of phospholipase in Bacteria associated with vaginitis

In this study, (90) high vaginal swabs were collected from women with abnormal vaginal discharge, who referred to Babylon Hospital for Maternal and Pediatrics, and Al-Hilla Teaching Hospital, during the period from November 2012 to March 2013 in Babylon Province. The patients’ ages ranged from (18–45 years). Seventy-six swabs were taken from non-pregnant women, and fourteen from pregnant women. Eighty-eight samples gave positive results of bacterial growth, and only two samples gave no growth of bacteria. In this study, the most common species of opportunistic bacterial isolates were Streptococcus agalactiae (28.4%), S. aureus (21.5%), Streptococcus mutans (15.9%), Coagulase-negative Staphylococcus (12.5%), Escherichia coli (7.9%), Acinetobacter (5.9%), Enterococcus (5.9%), Neisseria gonorrhoeae (2.2%). In this study, the effects of some antibiotics on bacterial isolates were investigated. It was shown that all isolates were sensitive to imipenem and to a lesser degree to Amoxi-Clav, amikacin, azithromycin, and ceftriaxone. However, some isolates showed resistance to one or more of the antibiotics used in this study, particularly to cefixime and ceftazidime. Production of extracellular phospholipase D in the presence of arachidonic acid was also detected. It was found that most bacterial isolates have the ability to produce this enzyme. Also, phospholipase extract was added to the culture media of Candida albicans, and it was found that there was an increase in the number of Candida (which was isolated in this study) after the addition of the enzyme extract. The study also included lecithinase detection; it was found that all vaginal isolates have the ability to produce this enzyme. Molecular detection of phospholipase (C) and its isozymes (A, B, C) was also studied by using specific polymerase chain reaction (PCR) primers. The results revealed that only one isolate of Escherichia coli and one isolate of Staphylococcus aureus gave positive amplicons for plcA, whereas only one isolate of Coagulase-negative Staphylococcus gave a positive amplicon for plcB, but none of the isolates gave positive results with plcC.

- تم في هذه الدراسة أخذ (90) مسحة مهبلية من النساء اللواتي يعانين من إفرازات مهبلية وراجعن مستشفى بابل للولادة والأطفال ومستشفى الحلة التعليمي، وذلك خلال الفترة من نوفمبر 2012 إلى مارس 2013 في محافظة بابل. تراوحت أعمارهن بين (18–45) سنة. تم أخذ (76) مسحة من غير الحوامل و(14) مسحة من الحوامل. (88) مسحة أعطت نتائج موجبة للنمو البكتيري و(2) فقط سالبة للنمو البكتيري. أظهرت النتائج أن أكثر أنواع البكتيريا الانتهازية المعزولة كانت Streptococcus agalactiae (28.4%)، تلتها S. aureus (21.5%)، ثم St. mutans (15.9%)، وCoagulase–negative staphylococcus (12.5%)، وE. coli (7.9%)، ثم Acinetobacter وEnterococcus (5.9%)، وأخيراً N. gonorrhoeae (2.2%). شملت هذه الدراسة أيضاً تأثير بعض المضادات الحيوية على العزلات البكتيرية، وأظهرت النتائج أن جميع العزلات كانت حساسة للإيميبينيم، وبدرجة أقل للأموكسيكلاف، والأميكاسين، والأزيثرومايسين، والسيفترياكسون. ومع ذلك، أظهرت النتائج مقاومة بعض العزلات لواحد أو أكثر من المضادات الحيوية المستخدمة في هذه الدراسة، وخاصة للسيفيكسيم والسيفتازيديم. كذلك تم الكشف عن إنتاج إنزيم الفوسفوليباز D خارج الخلية بوجود حمض الأراكيدونيك، وتبيّن أن معظم العزلات البكتيرية لها القدرة على إنتاج هذا الإنزيم. كما تمت إضافة مستخلص الفوسفوليباز إلى وسط الزراعة لفطر Candida albicans، ولوحظت زيادة في عدد خلايا Candida (التي عُزلت في هذه الدراسة) بعد إضافة مستخلص الإنزيم. وشملت هذه الدراسة أيضاً الكشف عن إنزيم الليسيتيناز، وتبيّن أن جميع العزلات المهبلية لها القدرة على إنتاج هذا الإنزيم. كما تم من خلال الدراسة الجزيئية الكشف عن إنزيم الفوسفوليباز (C) وأنزيماته الفرعية (A, B, C) باستخدام تقنية تفاعل البوليميراز المتسلسل (PCR). أظهرت النتائج أن عزلة واحدة فقط من E. coli وعزلة واحدة من S. aureus أعطت نتيجة موجبة لجين plcA، في حين أن عزلة واحدة فقط من Coagulase-negative staphylococcus أعطت نتيجة موجبة لجين plcB، بينما لم تُظهر أي من العزلات نتائج موجبة لجين plcC.
